The castles are pretty repetitive so I would say choose one between Osaka and Himeji, I heard Himeji is better but I've never visited there.
Drop Osaka Castle and travel on to Himeji instead. Much prettier and less of a museum-feel inside.
If you girls want to see a really nice (and genuine) castle, take a train to Himeji instead. Absolutely beautiful.
Himeji is the most imposing of the castles and has some superb aesthetics and surrounding infrastructure. It is definitely a must see on any trip to Japan. But, because of all this, it is absolutely packed to the brim with tourists. You are shuffled along at a quick pace from section to section, and it’s hard to really stop and appreciate its majesty. It’s both essential and a bit exhausting.
Hime-ji is wonderful, do the combo ticket castle + botanical garden.
Also if you do Himeji it's much better than Osaka Castle.

Himeji Castle is very much a defensive castle, not a palace that is ornately decorated. My GF was very disappointed, but I thought it was pretty cool.
Himeji Castle is probably 90 minutes, maybe up to 2 hours depending on your pace. It's also a decent walk to/from the train station, so factor that in as well.
Himiji also do Mt Shosa and be at the castle when it opens 9:00 as it gets crowded quick.
